My brother's Haplogroup is U, while mine is U2. I have one more marker than he and it's 16051G. How does this happen? I am younger than he.

Not sure what you mean by full name of test. We both had HVR1 and HVR2 with FTDNA. He tested U and at that time there were other folks with U2 that matched him in all but the 16051G, which I have. I just got my results. That is our only difference.

Both are mtDNAplus which are not the complete mtDNA only a subset. I'm not sure that name is exact same test considering the difference is 8 years. It's also possible U2 marker was inconclusive on the older test.
Hopeful Rebekah knows or can check with FTDNA to answer your question. if not you can call FTDNA support and they will be able give you a definite answer.

Back when he was first tested, his Haplogroup was U*, which was later changed to U. There were some matches that I am now matched with, but he had the one difference was this same marker. When I inquired at that time, I was told that he probably had a back mutation. So, when I decided to go ahead and have my dna tested, I was surprised to find that I had that marker.
